You can't get rid of the number behind your nick in specator chat.
The Players can't see spectator chat.
By setting your spectatorname you also set your in game nickname. No further changes needed.

The number is indeed which number spectator you are. Don't think there is any way you can get rid of it, but can't really see a reason why you would need too anyway.
1) Only other spectators can read your comments but the players can hear when you use one of the available emotes (/hurt, /cheer, /boo).
2) As far as I know <red> will change to the name you enter as "spectatorname=", <blu> will be your opponents name if they have updated their options.nfo in the same way.
